AFAIK, 2.1.3 is the still the only *supported* version of Python for
running Zope, including the head;  I don't know of specific problems
with 2.2.1, but believe some in the community have reported stability
issues with it when running for longer periods, at least on some
platforms.  Can anyone on zope-coders confirm or deny?

BTW, I would be experimenting with 2.2.1 right now on a client
gig if I could get the libxml2-python and libxslt-python stuff to build
properly under that version.
